Does the operation of Overhead Electricity Operated Crane (EOT crane) has any regulation requiring mandatory installation of operational siren (Alarm).

If it is a requirement (not best practices), please can someone clarify and point to relevant regulations.

I am not aware of any regulation which requires the mandatory use of an audible alarm. The cranes I have seen in operation all use an alarm when being operated. I suspect the manufacturer's operation manual is the place where you mind find a reference to an audible alarm, in which case it would be adhering to the manufacurer's instructions. Ultimately it could come down to your RA, assuming you have one in place, what does that say about the controls for the safe operation of an Overhead Electricity Operated Crane?

What would the purpose of the alarm be?

BS EN 15011:2011+A1:2014 Which is the British Standard for Bridge and gantry cranes states: 

5.5.2.1 Rated capacity indicator Rated capacity indicators in accordance with EN 12077-2 shall be provided on bridge and gantry cranes where the rated capacity varies with the position of the load. Such indicators shall give a visual warning at 90 % of the rated capacity and a visual or audible warning at overload.
