"Have you any reservations about my suitability to do the job you want filled?"
and be prepared to respond constructively (not defensively) to candid replies.

If face-to-face VALID feedback that you ask for 'sets you back', you may well not be suitable for a challenging vacancy.
As a registered coaching and occupational psychologist, I have explicitly encouraged thousands of candidates who ask your question to invite intelligent interviewers for feedback candidly. If it does 'set you back', it's up to you to correct any misunderstandings or misperceptions.
Not all interviewers are honest, however, and may not tell the truth, even if they profess publicly and loudly to do so, with bells on.
When I was interviewed for Fellowship of another chartered professional society and asked this question, I was informed, 'People here know much more about.... than you do!'; it was straightforward for me to reply, 'I think that's because.....'. To which the person who answered the question said, 'Well, I'm going to recommend you for the Fellowship'. He did and I've been a Fellow of the CIPD for over 20 years now.
But DO NOT ask if interviewers have any reservations about your suitability UNLESS you actually do want to learn what they think. Repeat, DO NOT do so unless you want to learn about how you are perceived by interviewers if you are unwilling to listen and learn from what they say.

KieranD I would have no problem asking that question, would listen to there answer and give my own opinion why I would bet a great fit for there role.
